Acridine Orange is a reddish-brown fluorescent powder, which has a high affinity in binding nucleic acids. It readily enters cells and gives green fluorescence in presence of the double stranded DNA and gives red fluorescence in presence of single stranded DNA or RNA. DNA and RNA have a maximum excitation and emission at 502 and 525 nm and 460 and 650 nm respectively. Acridine Orange is known as lysosomal stain, a positive solvatochromic stain as well.
